10X Single-Cell Sequencing and Visium CytAssist Spatial Gene Expression

 

 

Single Cell Sequencing

Single Cell RNA sequencing at our facility is done through the 10X Genomics platform which offers multiple different assay options for uncovering gene expression in unique or rare cell populations within a heterogeneous sample, under various conditions and cell states. The Chromium X Controller uses Next GEM technology that allows thousands of cells per run to be individually barcoded before downstream library construction. Once sequenced with the Illumina NextSeq2000, reads can then be mapped back to individual cells via their unique barcode.

10X Assays offered include:

  • 3’ gene expression
  • 5’ gene expression
  • Immune profiling (TCR and/or BCR)
  • ATAC sequencing
  • Fixed RNA Profiling (cells or nuclei)

High viability cells are required for most assays.

Visium CytAssist Spatial Gene Expression

With the 10X Visium CytAssist, spatial transcriptomics can be performed on human or mouse tissue slides. The CytAssist instrument facilitates the transfer of transcriptomic probes from standard glass tissue slides to Visium barcoded slides, enabling whole transcriptome spatial profiling. Capture areas are either 6.5mm x 6.5mm or 11mm x 11mm. Assays are compatible with H&E or immunofluorescence stained FFPE or fresh frozen tissue sections.

We offer standard Visium library preparation with 5000 barcoded spots per capture area or Visium HD library preparation with single-cell scale resolution.
